I'd start with the basics and just do a normal webforms or mvc asp.net site. Once you understand the basics it will then become clearer to you why and when you'd use things like ajax, or bootstrap, and can then try sites with those too. You probably don't want to try and learn too much at one time, spend time getting a good understanding of the fundamentals and debugging.
