I think that this issue can be related with system folder SysWOW64 that you trying access. Mmm, not you but ASP.Net application, that, of course, has not any permissions on this folder.

So try copy your file in another folder, for example: C:\temp, and try accessing from there.
And make sure that this file is not currntly open by other applications.
