I can easily create multiple columns in a combobox using sample data. Question is, how can I create one manually, without using sample data?

You can edit the sample data in the code editor by opening the SampleData folder and finding the xaml document for your particular source. Items can be edited there.

You need to edit the template to create columns.  For example, create your combobox, right click it, select edit additional templates, item template, create empty.  Now you will be editing the template for each item. You could do this with your sample data version first to get familiar with how things should look, and how the data bindings should be set up.
