Can any one help in restricting access on _layouts/MySubs.aspx.

I don't want user to open this URL to Add or Delete alerts.

You can activate the feature called "ViewFormPagesLockDown Feature" on your site collection. So the user group with restricted read permission cannot access the _layout page in the site collection.
