We are currently on TFS2015 update 3 and relying heavily on the TFS2013 Test Controllers and Agents as our users really need the ability to be able to select a single test
case in MTM and just run the automated test case associated to it.
As the moment we're looking at rolling out the test automation framework on a large scale for our enterprise test teams and they’re concerned about the load balancing capability of the Test Controller / Test Agent framework. What I’m specifically interested in is what is the recommendation from Microsoft regarding how many Test Agents a single Test Controller should have registered against it and are there any hard limits on this ?

There is no limits for the number of test agent. You can install a test controller on any suitable computer, and a test agent on each of the computers that are running components of the system under test.
Lab tests. You want to run tests either manually or automatically on a lab environment by using Microsoft Test Manager and Visual Studio Lab Management. In this case you have to:
Install a test controller on a suitable machine.
Register the test controller with a Team Foundation project.
Install a test agent on each computer or virtual machine template on which tests will be run.
