I want to keep vb.net app exe and sql database files in a readable/writable cd or in a pen drive
vb.net app will access,store and update data in sql database.Dvd or pen drive will hold data related to that application.Client PC will have sql server software.
Finally SQL files in DVD or pen drive should have updated data.

Is there any way to do this?

You also have to understand that force a client to install SQL server is insane - SQL server is a huge bit of software and not meant to sit on client's computer...
The best thing you can do is using SQL Compact to embed the database into your application (not physically but logically)...
