In Expression Encoder 4, is it possible to erase the audio associated with a video clip, and then replace the audio with something else?  If yes, how is that done?  I have EE4 SP2 installed on Windows 7 x64.

Yes you can import the video and then go to the Enhance tab and find the Audio section in the Pre-Processing part. There is a control to turn the volume down to 0 (for the audio on the video file).

Now a little further down on the that screen you can see the audio overlay section. Click on the checkbox and then you can add the audio file that you want to use for the audio when you encode.
