I would like to know if the transformation itinerary services (both messaging and orchestration) of the toolkit benefit from the BizTalk large message transformation capability (the different transformation engine which is used when transforming large messages so the message data is buffered to the file system instead of being loaded into memory)?

The current message transform uses a memory stream for the resulting message, but should use the BizTalk VirtualStream which gives the behavior you would want for a large message.   So the answer is no for the message transform service. The orchestration uses this capability: http://msdn.microsoft.com/en-us/library/aa950573(BTS.10).aspx.
