I'm trying to put together a SharePoint list that would enable users to select a timeslot from a Lookup list but don't want multiple people to be able to select the same timeslot. So it would be good to remove it from the displayed available timeslots?

I would make one list (Calendar list), named Appointments.  I would use an approval workflow (Built-in or Custom). Once the request for the appointment time has been approved there will be a field specifying that. The view that people would be directed to, would have those dates populated.  If you're using a list, then the view they go to would have a filter to only show rows without an approved/requested entry.
