I would like to change the document into read-only access mode when the user start the approval workflow.

Because if the requester changed the document content, then the approval has no meaning.

For your requirement, assuming you are using SharePoint 2010 platform workflow, before the step which contains"Start Approval Process" action, add an Impersonation Step and insert a "Replace List Item Permissions" to remove the permissions on the user that you don't want to have, like the below:
