I have just set up a couple of SharePoint 2016 servers. One is WFE+DC and the other is Application and Search. I have configured search for the SharePoint site and also a people search of mysites. The crawl account has read access and I've given it the "Manage auditing and security log" role on the file server. I can login on the search server with this account and happily access the file share so it does have access. What's more I still have the old SharePoint 2013 server which uses exactly the same account to crawl exactly the same file server using exactly the same URL and it works happily. How can it be that it works with SharePoint 2013 but doesn't work with SharePoint 2016?

Like you said that you also got the error message if you accessed it directly, this issue should be about permissions on DFS server.
