I was wondering if there is a way to edit permissions on a sharepoint list so only a user given the permission to can edit a single column in a list.

So basically my HR manager would like to put the annual leave form in a sharepoint list, which is fine. However 2 columns in that list she only wants herself to be able to edit. is this possible?

SharePoint doesn't support custom column permissions out of the box.  There have been attempts to do it on the third party market, but the problem is that users have the ability to add any column to a personal view, which would allow them to see that column.  Its not really possible to restrict that.
