We have 2 application servers using ASR. These are configured as websphere cluster for application load balancing.

Want to know, what happens post faiover to Azure. Do I need to re-configure the websphere cluster?

That depends on how you have networking and compute configured. As well as access requirements.

For example, your DNS service will point to the VMs in their current location, you will need a step to re-point to the Azure location.
