I guess you are asking how to upgrade your existing BizTalk 2016 standard edition to Enterprise?

If possible, because now the servers are usually virtual so it's easy to spin up a new environment, just build a new production environment with Enterprise version installed and migrate your code. Why take chances.

However, if it is not possible then maybe you can take the required backups and reinstall the enterprise version installer. 

• Back-up of databases
• BizTalk application by MSIs.
• Backup the binding, parties, policies, vocabularies or any artefact related to BuzTalk.
• BTSNTSVC.config file.
• Backup the Enterprise Single Sign-On secret

Also, please make sure after the upgrade you also install the latest cumulative upgrade or Feature Pack 3 just to keep things up to date.
