I have users using a desktop in a cloud service, this is not a Microsoft one but it is hosted in Azure.  The region is UK South and all users access this remotely.

I am looking to add a core file server (or Azure Files) into Azure to support a new service however I must put this stored data in an alternate region to the one the desktop service exists in. This is for compliance purposes.

I read that if the file share and VM are in the same Azure region, there is no additional charge for the traffic between the file share and the VM however if the file share and the VM are in different regions, the traffic between them are charged as external bandwidth.

If my users are accessing from a virtual host in one region accessing and storing data in another, this seems likely to incur significant cost. Is there any way to work out what this might be based on say user and expected file sizes per daily use?

You would want to set up a test environment, measure the resulting traffic, and extrapolate the results to what you'd expect in the production environment.
