One of the requirement in current project is to make the collection read only. Collection almost has 70+ projects hence I want to make the collection itself as read only. Plus I am not aware how to make project read only as well. However making collection read only makes more sense to me instead of making each project read only and then re-doing it.

you can't set a Team Project resp. a TFS Collection to readonly.

the only way to disable modification is to remove members of the groups with modify permissions.
